质膜H+ATP酶艾美捷应用实例:
图:来自拟南芥(1),大麦(2),玉米(3),烟草质膜部分的20μg总蛋白,2.5μg(4) ,用蛋白质提取缓冲液,PEB(AS08 300,用3至5体积的均质缓冲液),在70°C下变性10分钟,并在4-12%NuPage(Invitrogen)LDS-PAGE上分离,并印迹1h至PVDF。在20mm Tris,137mm氯化钠ph7.6,0.1%(v/v)的封闭试剂中转移后立即封闭斑点Tween-20(TBS-T)在室温下搅拌1小时。Blots在一抗体中以1:5000的稀释度在室温下孵育1h温度搅拌。倾析出抗体溶液并将印迹短暂冲洗两次,然后洗涤一次15分钟和3次在室温下在TBS-T中搅拌5分钟。用二抗体(抗兔IgG辣根过氧化物酶)孵育小鼠偶联的推荐二抗AS09 602)在2%封闭溶液中于室温搅拌稀释至1:20 000,持续1h。如上洗涤印迹,并根据制造商的说明用化学发光检测试剂显色5分钟。使用CCD成像仪(FluorSMax,Bio-Rad)和Quantity One软件(Bio-Rad)获得印迹的图像。曝光时间为2分钟。
