背景资料Plectin is a prominent member of an important family of structurally ａnd in part functionally related proteins, termed plakins or cytolinkers, that are capable of interlinking different elements of the cytoskeleton. Plakins, with their multi-domain structure ａnd enormous size, not only play crucial roles in maintaining cell ａnd tissue integrity ａnd orchestrating dynamic changes in cytoarchitecture ａnd cell shape, but also serve as scaffolding platforms for the assembly, positioning, ａnd regulation of signaling complexes (reviewed in PMID: 9701547, 11854008, ａnd 17499243). Plectin is expressed as several protein isoforms in a wide range of cell types ａnd tissues from a single gene located on chromosome 8 in humans (PMID: 8633055, 8698233). Until 2010, this locus was named plectin 1 (symbol PLEC1 in human; Plec1 in mouse ａnd rat) ａnd the gene product had been referred to as "hemidesmosomal protein 1" or "plectin 1, intermediate filament binding 500kDa". These names were superseded by plectin. The plectin gene locus in mouse on chromosome 15 has been analyzed in detail (PMID: 10556294, 14559777), revealing a genomic exon-intron organization with well over 40 exons spanning over 62 kb ａnd an unusual 5＇ transcript complexity of plectin isoforms. Eleven exons (1-1j) have been identified that alternatively splice directly into a common exon 2 which is the first exon to encode plectin＇s highly conserved actin binding domain (ABD). Three additional exons (-1, 0a, ａnd 0) splice into an alternative first coding exon (1c), ａnd two additional exons (2alpha ａnd 3alpha) are optionally spliced within the exons encoding the acting binding domain (exons 2-8). Analysis of the human locus has identified eight of the eleven alternative 5＇ exons found in mouse ａnd rat (PMID: 14672974); exons 1i, 1j ａnd 1h have not been confirmed in human. Furthermore, isoforms lacking the central rod domain encoded by exon 31 have been detected in mouse (PMID:10556294), rat (PMID: 9177781), ａnd human (PMID: 11441066, 10780662, 20052759). The short alternative amino-terminal sequences encoded by the different first exons direct the targeting of the various isoforms to distinct subcellular locations (PMID: 14559777). As the expression of specific plectin isoforms was found to be dependent on cell type (tissue) ａnd stage of development (PMID: 10556294, 12542521, 17389230) it appears that each cell type (tissue) contains a unique set (proportion ａnd composition) of plectin isoforms, as if custom-made for specific requirements of the particular cells. Concordantly, individual isoforms were found to carry out distinct ａnd specific functions (PMID: 14559777, 12542521, 18541706). In 1996, a number of groups reported that patients suffering from epidermolysis bullosa simplex with muscular dystrophy (EBS-MD) lacked plectin expression in skin ａnd muscle tissues due to defects in the plectin gene (PMID: 8698233, 8941634, 8636409, 8894687, 8696340). Two other subtypes of plectin-related EBS have been described: EBS-pyloric atresia (PA) ａnd EBS-Ogna. For reviews of plectin-related diseases see PMID: 15810881, 19945614. Mutations in the plectin gene related to human diseases should be named based on the position in NM_000445 (variant 1, isoform 1c), unless the mutation is located within one of the other alternative first exons, in which case the position in the respective Reference Sequence should be used.

抗体保存得当与否直接决定了抗体的活性和使用效果。如果抗体保存得当，大部分抗体活性都可以维持数月甚至数年。请按照说明书或者抗体标签上推荐的保存条件正确保存抗体！无论是保存还是运输，绝对避免反复冻融抗体！

1.拿到抗体后请务必在1000-3000转离心1-2分钟后再打开管盖进行分装和保存！（由于运输过程中反复颠簸，管内微量抗体容易聚集在管盖处，一旦直接打开容易流失抗体。请放心抗体出厂时保证已装入足量。）

2.对于大部分抗体，比较合适的保存方式是分装后保存在-20℃冰箱。

◇分装可以程度降低反复冻融对抗体活性的损害，Plectin抗体同时也降低了多次从同一管中吸取抗体造成污染的可能性。

◇分装的量以一次实验用完为好，但建议*少不要少于10μL每份。分装体积越小，抗体浓度可能会受到蒸发以及管壁吸附的影响，同时分装次数越多移液吸头吸附的抗体也越多，可能会误认为抗体没有足量装入。（用无菌的进口0.2ML离心管来分装抗体，国产的小管一般没有经过很好的硅化处理，容易吸附抗体蛋白，影响抗体活性。）

◇分装时请将无菌的微量移液器吸头先插入管底部反复吹打几次后再吸出抗体，Plectin抗体以便抗体有效成分与抗体保护剂充分混匀。

◇复融后的分装抗体如果一次用不完，请将剩余母液保存在4℃冰箱，避免再冻起来！

◇绝对避免将抗体保存在自动除霜冰箱的冷藏室中。尽量将抗体保存在手动除霜冰箱里面，而不是在冰箱门上。

3.大部分抗体收到后4℃短暂保存1-2周对抗体活性是基本没有影响的。如果抗体很快（1-2周内）就能使用完，推荐在4℃保存，避免反复冻融对抗体活性的损害。如果要长期保存则分装后-20℃保存。

4.即用型抗体请务必保存在4℃冰箱，一定不能冷冻保存。

5.特殊抗体的保存：

◇酶联抗体：一般保存在4℃，尽量避免冷冻起来。否则可能会导致酶活力的下降或者丧失。

◇偶联抗体：所有偶联抗体需要4℃避光保存。尤其是荧光标记抗体，对光极为敏感，因此所有实验阶段也是要避光操作的。
