保存条件  Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month ａnd for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.

 Methylation at the 5＇-position of cytosine is the only known naturally occurring covalent modification of the mammalian genome. DNA methylation requires the enzymatic activity of DNA 5-cytosine methyltransferase (Dnmt) proteins, which catalyze the transfer of a methyl group from S-adenosyl methionine to the 5＇-position of cytosines, thereby repressing expression of the target gene. Dnmt3L (DNA (cytosine-5)-methyltransferase 3-like) is a 387 amino acid protein that contains one ADD-type zinc finger ａnd is a member of the Dnmt family. Localized to the nucleus ａnd expressed at lows levels in thymus, testis ａnd ovary, Dnmt3L does not exhibit DNA methyltransferase activity, but is able to stimulate de novo methylation by Dnmt3 ａnd is thought to play a key role in the establishment of genomic imprints. Additionally, Dnmt3L interacts with histone deacetylase 1 (HDAC1) and, through this interaction, mediates transcriptional repression. Multiple isoforms of Dnmt3L exist due to alternative splicing events.

Function : Catalytically inactive regulatory factor of DNA methyltransferases. It is essential for the function of DNMT3A ａnd DNMT3B. Activates DNMT3A ａnd DNMT3B by binding to their catalytic domain. Accelerates the binding of DNA ａnd AdoMet to the methyltransferases ａnd dissociates from the complex after DNA binding to the methyltransferases. Recognizes unmethylated histone H3 lysine 4 (H3K4) ａnd induces de novo DNA methylation by recruitment or activation of DNMT3.

Subunit : Homodimer. Heterotetramer composed of 1 DNMT3A homodimer ａnd 2 DNMT3L subunits (DNMT3L-DNMT3A-DNMT3A-DNMT3L). Interacts with histone H3 (via N-terminus).

Subcellular Location : Nucleus.
Tissue Specificity : Expressed at low levels in several tissues including testis, ovary, ａnd thymus.
Similarity : Belongs to the C5-methyltransferase family.
Contains 1 ADD-type zinc finger.
