小鼠神经特异性烯醇化酶(NSE)elisa试剂盒*新闻快讯标本的处理:
(1)细胞培养上清液
根据需要用试剂盒中的标准稀释液稀释样品,然后按照说明书所述方法检测。推荐稀释浓度是稀释5倍。
(2)脑脊液
根据需要用试剂盒中的标准稀释液稀释样品,然后按照说明书所述方法检测。推荐稀释浓度是5倍。
(3)血浆
①用EDTA2K离心收集在真空血液收集管中的血液,5000g,15分钟,4℃,分离血浆样品,然后储存在零下80℃直到使用。
②用试剂盒中的标准稀释液5倍稀释血浆以避免血浆中的干扰物质影响检测,按照说明书指定方法检测。
酶标比色仪简称酶标仪,通常指专用于测读ELISA结果吸光度的光度计。针对固相载体形式的不同,各有特制的适用于板、珠和小试管的设计。许多试剂公司配套供应酶标仪。
小鼠神经特异性烯醇化酶(NSE)ELISA试剂盒*新闻快讯酶标仪的主要性能指标有:测读速度、读数的准确性、重复性、精确度和可测范围、线性等等。优良的酶标仪的读数一般可精确到0.001,准确性为+-1%,重复性达0.5%。举例说,若某孔测得的A值为1.083,则该孔相对于空气的真实A值应为1.083+-0.01(1.073~1.093),重复测定数次,其A值均应1.083+-0.05(1.078~1.088)在之间。酶标仪的可测范围视各酶标仪的性能而不同。普通的酶标仪在0.000~2.000,新型号的酶标仪上限拓宽达2.900,甚至更高。超出可测上限的A值常以“*”;或“over”或其它符号表示。应注意可测范围与线性范围的不同,线性范围常小于可测范围,比如某一酶标仪的可测范围为0.000~2.900,而其线性范围仅0.000~2.000,这在定量ELISA中制作标准曲线时应予注意。
