血清保存方法
血清是指血液凝固后,在血浆中除去纤维蛋白分离出的淡黄色透明液体或指纤维蛋白已被除去的血浆。其主要作用是提供基本营养物质、提供激素和各种生长因子、提供结合蛋白、提供促接触和伸展因子使细胞贴壁免受机械损伤、对培养中的细胞起到某些保护作用。那么血清应该如何进行保存呢?
(1)需要长期保存的血清必须储存于-20C - 70°C 低温冰箱中。4C冰箱中保存时间切勿超过1个月。由于血清结冰时体积会增加约10%因此,血清在冻入低温冰箱前,必须预留一定体积空间,否则易发生污染或玻璃瓶冻裂。
(2)一般厂商提供的血清为无菌,无需再过滤除菌。如发现血清有悬浮物,则可将血清加入培养液内一起过滤,切勿直接过滤血清。
(3)瓶装血清解冻需采用逐步解冻法:-20C至-70C低温冰箱中的血清放入4'C冰箱中溶解1天.然后移入室温,待全部溶解后再分装.在溶解过程中需不.断轻轻摇晃均匀(小心勿造成气泡),使温度与成分均一,减少沉淀的发生.切勿直接将血清从-20°C进入37'C解冻,这样因温度改变太大,容易造成蛋白质凝集而出现沉淀。
(4)热灭活是指56C,30分钟加热已完全解冻的血清.加热过程中须规则摇晃均匀.此热处理的目的是使血清中的补体成分灭活.除非必须,一般不建议作此热处理,因为热处理会造成血清沉淀物显著增多,而且还会影响血清的质量.补体参与反应有:细胞毒作用,平滑肌细胞收缩,肥大细胞和血小板释放组胺,增强吞噬作用,促进淋巴细胞和巨噬细胞发生化学趋化和活化。
(5)切勿将血清在37C放置太久,否则血清会变得浑浊,同时血清中的有效成分会破会而影响血清质量。
(6)血清中的沉淀物
絮状物:主要是血清中的脂蛋白变性及解冻后血清中纤维蛋白造成,这些絮状物不会影响血清本身的质量,可用离心3000rom,5分钟去除,也可不用处理。

(2) the serum provided by the general manufacturer is aseptic and does not need to be filtered and sterilized. If the serum is found to have suspended matter, the serum can be added to the culture medium to filter together, do not filter the serum directly.
(3) the thawing of bottled serum should be done step by step: the serum from-20C to-70C cryogenic refrigerator should be dissolved in 4 / C refrigerator for 1 day. Then move to room temperature and divide it up until it is completely dissolved. In the process of dissolution, whether or not. Break gently shake evenly (careful not to cause bubbles), make the temperature and composition uniform, reduce the occurrence of precipitation. Do not thaw the serum directly from-20 °C to 37 °C, so that because the temperature changes too much, it is easy to cause protein aggregation and precipitation.
(4) Thermal inactivation refers to 56C, heating completely thawed serum for 30 minutes. It is necessary to shake evenly in the heating process. The purpose of this heat treatment is to inactivate complement components in serum. Unless necessary, this heat treatment is not generally recommended, because heat treatment can cause a significant increase in serum precipitates and affect the quality of serum. Complement participation includes cytotoxicity, contraction of smooth muscle cells, release of histamine from mastocytes and platelets, enhancement of phagocytosis, and promotion of chemical chemotaxis and activation of lymphocytes and macrophages.
(5) do not place the serum at 37C for too long, otherwise the serum will become turbid and the active components in the serum will break and affect the quality of the serum.
(6) sediments in serum
Flocs: mainly caused by the denaturation of lipoprotein in serum and fibrin in serum after thawing, these flocs will not affect the quality of serum itself, can be centrifuged 3000 rom, removed for 5 minutes, or can not be treated.
