Parkinson＇s disease (PD) is one of the most common degenerative diseases of central nervous system in middle-aged ａnd elderly people, which has the characteristics of recessive onset ａnd slow progress. With the acceleration of population aging in China, the incidence of PD is increasing day by day. At present, there are about 2 million PD patients in China, ａnd about 100,000 new PD patients are added every year. The prevalence rate of people over 55 years old is 1.07%, ａnd that of people over 75 years old is 2.5%. It is estimated that the number of PD patients in China will be nearly 5 million by 2030. The main pathological feature of PD is the progressive degeneration ａnd decrease of dopaminergic neurons in the substantia nigra compacta of midbrain, ａnd its mechanism is closely related to the enhancement of oxidative stress ａnd mitochondrial dysfunction. When PD dyskinesia appears clinically, the number of dopamine,DA neurons in substantia nigra has decreased by more than 50%, ａnd the content of DA in striatum has decreased by more than 80%. At present, there is no fundamental treatment, so it is of great clinical significance to carry out neuroprotective treatment as soon as possible. 1. 6-OHDA is a dopamine analogue, its structure is similar to dopamine, ａnd it is often mistaken as DA neurotransmitter to ingest DA neurons, which selectively destroys DA neurons ａnd causes them to die. 6-OHDA was detected in PD patients＇ brains ａnd urine samples. As a selective toxin of dopamine neurons, 6-OHDA is often used to make animal ａnd cell models of Parkinson＇s disease. Effects of uric acid ａnd 6-OHDA on PC12 cell viability objective: to evaluate the effects of uric acid UA ａnd 6-OHDA on PC12 cell viability. Methods: PC12 cells were cultured with DMEM containing 10% calf serum, ａnd the solution was changed for 2 ~ 3 days. Uric acid was divided into control group (DMEM), 100,200,300,400 ａnd 500μmol/L uric acid groups. 2. Uric acid reduces the toxicity of 6- hydroxydopamine to PC12 cells Objective: To evaluate the effect of uric acid on reducing the toxicity of 6-OHDA to PC12 cells.
