| comments: | the proteins were isolated from various tissues by preparing a tissue homogenate in the presence of protease inhibitors. protein samples (75 µg) from each tissue were solubilized in sds-lysis buffer and electrophoresed in a 10 well, 4-20% sds-polyacrylamide gradient gel, followed by electroblotting on pvdf membrane. components one protein blot of normal human tissue samples arranged as follows: lane 1: protein marker lane 2: human liver lane 3: human brain lane 4: human lung lane 5: human kidney lane 6: human spleen lane 7: human testis lane 8: human ovary lane 9: human heart lane 10: human pancreas the protein molecular weight markers are as follows: myosin: 203,000 β-galactosidase: 120,000 bovine serum albumin: 90,000 ovalbumin: 51,700 carbonic anhydrase: 34,100 soybean trypsin inhibitor: 28,000 lysozyme: 20,000 aprotinin: 6400 remove the blot from the pouch and wash with an appropriate buffer (1x tbst or pbst) 1-2 times. block the membrane with a protein blocking agent and incubate with the primary and secondary antibodies diluted in blocking solution. develop the blot with chemiluminescent or chromogenic detection reagents for the detection of the specific protein |
