application data: | tmb (soluble substrate) produces a higher signal with the lowest background and outperforms other commercially available tmb single-component substrates offering the best signal to noise ratio possible. different lots of tmb (soluble substrate) were compared in an elisa containing 5 µg horseradish peroxidase (hrp) conjugate. the absorbance at 620 nm was measured at the indicated times. tmb (soluble substrate) was stored at 0°c, 4°c, 25°c or 37°c for 30 days and then tested for stability. the following graph demonstrates the stability of tmb (soluble substrate) where performance and background absorbance values remain consistent even after 30 days of stress conditions. |

background: | tmb (soluble substrate) is a non-carcinogenic chromophore for use in elisa and other microwell applications. tmb (soluble substrate) contains 3,3",5,5"-tetramethylbenzidine formulated in a mildly acidic buffer with proprietary stabilizers. it is supplied as a one component liquid solution at a ready-to-use working dilution for application use. unreacted substrate should be colorless or a very light-bluish-green prior to use. the substrate system develops a blue reaction product when reacted with peroxidase conjugates in microwell applications for visual reading at a wavelength of 370 nm and between 620 and 650 nm. for endpoint assays, the addition of an equal volume of 1n or 2n hcl, or 1n h2so4 stops the reaction and turns the blue reaction product to a yellow reaction product and is read at a wavelength of 450 nm. endpoint assays should be read between thirty and sixty minutes after addition of stopping solution. tmb (soluble substrate) produces a soluble reaction product that is not recommended for membrane or immunohistochemical applications, which require a precipitating reaction product. |
