BackgroundInformation
Flagtag（alsoknownasDDDDKtagorAnti-Dtag）isapolypeptideproteintagthatcanbeaddedtoaproteinusingRecombinantDNAtechnology.Itcanbeusedforaffinitychromatography,thenusedtoseparaterecombinant,overexpressedproteinfromwild-typeproteinexpressedbythehostorganism.Itcanalsobeusedintheisolationofproteincomplexeswithmultiplesubunits.ThepeptidesequenceoftheFlagtagisasfollows:N-DYKDDDDK-C（1012Da）.Itcanbeusedinconjunctionwithotheraffinitytagsforexampleapolyhistidinetag（His-tag）,HA-tagormyc-tag.ItcanbefusedtotheC-terminusortheN-terminusofaprotein.AFlagtagcanbeusedinmanydifferentassaysthatrequirerecognitionbyanantibody.Flagantibodiesrecognizetheepitopeincertainpositions,e.g.exclusivelyN-terminalorposition-insensitive.
Application Notes
Optimal working dilutions should be determined experimentally by the investigator. Prepare working dilution immediately before use. Suggested starting dilutions are as follows: Western Blot （1:1000-1:10,000）, Immunoprecipitation ａnd Immunofluorescence or Immunocytochemistry （1:200-1:800）.
