Description | Plasma fibronectin level is elevated in severe coronary artery disease. Increased plasma fibronectin levels are related with venous thromboembolism (VTE) particularly in males, and extend the probable association between biomarkers and risk factors for arterial atherothrombosis and VTE. Fibronectin plays a role in several cellular processes, including tissue repair, embryogenesis, blood clotting, and cell migration/adhesion. Fibronectin consists in two main forms: 1) as an insoluble glycoprotein dimer that serves as a linker in the etracellular matrix and 2) as a soluble disulphide linked dimer found in the plasma. The plasma form is produced by hepatocytes, and the ECM form is synthesized by fibroblasts, chondrocytes, endothelial cells, macrophages, as well as certain epithelial cells. Fibronectin alos takes part as a general cell adhesion molecule by anchoring cells to collagen or proteoglycan substrates. Fibronectin organizes cellular interaction with the ECM by binding to different components of the extracellular matrix and to membrane-bound Fibronectin receptors on cell surfaces. Solubility | Purified fibronectin has a tendency to form insoluble aggregates upon reconstitution. We suggest to reconstittue the 1mg Fibronectin with a chaotropic agent such as urea at room temperature at a concentration of 0.2mg/ml using sterile water. Let stand 1-2 hours. |
Stability | Lyophilized Fibronectin although stable at room temperature for 1 week, should be stored desiccated below -18°C. Upon reconstitution Fibronectin should be stored at 4°C between 2-7 days and for future use below -18°C. Please prevent freeze-thaw cycles. |

Cell Culture Application | Fibronectin is useful for the induction of cell attachment to a variety of surfaces including plastic and glass tissue culture labware, petri dishes, coverslips, microcarrier beads, etc. Fibronectin is useful for growth and maintenance of cells in low serum conditions. In general, the reconstituted fibronectin should be diluted with sterile physiological saline or serum-free medium to a concentration of 10-50ug/ml. The plates are treated with the solution, at room temperature 1-2 hours, the supernatant removed and the plates rinsed with fresh media or buffer. Growth media is then added and the coated plates seeded with a suspension of freshly harvested cells. Care should be taken to inactivate serum proteases used in the preparation of the cells, since fibronectin is very sensitive to proteolysis. Optimum concentration of fibronectin depends on the types of cell and the particular surface used. Generally, use 1-5ug/cm2 of growth surface. |

溶解性:
纯化的纤维连接蛋白在重悬后很容易形成不溶性聚合物。我们建议使用离液序列高的试剂如尿素,在室温下重悬1mg的纤维连接蛋白,使其浓度达到0.2mg/ml。
静置1-2小时。
稳定性:
冻干的淋巴细胞分离液虽然在室温可以贮存1星期,贮存在-18°C以下。
重悬的纤维连接蛋白4°C可以保存2-7天。保存要贮存在-18°C以下。
不要反复冻融。
纯度:
SDS-PAGE检测,纯度大于95.0% 。
细胞培养应用:
纤维连接蛋白在诱导细胞黏附到各种表面包括塑料,玻璃组织培养器皿,培养皿,盖玻片,微载体磁珠,等非常有用。纤维连接蛋白能够使细胞在低血清培养基中生长。一般来说,应该用无菌生理盐水或无血清培养基重悬纤维连接蛋白,使稀释的浓度保持在10-50ug/ml。用溶液室温处理培养皿1-2小时,然后弃掉液体,用新鲜培养基或者缓冲液冲洗培养皿。加入生长培养基,然后在包被的培养皿中接种新鲜的细胞悬液。应注意要在培养的细胞中加入灭活的血清蛋白酶,因为纤维连接蛋白很容易水解。的纤维连接蛋白浓度取决于细胞类型和使用表面积。一般来说,使用生长面积为1-5ug/cm2。
用法:
