醛化红细胞特点:
1.醛化红细胞镜检时一般呈两面凸起的园盘状,散在,不聚集成团。
2.红细胞醛化后沉积细胞体 积约增加20%,红细胞固有形态无变化,呈棕色,在有Na+存在时颜色略红。
3.醛化细胞具有较强的抗破坏能力,反复冻融,加热至60,蒸馏水中低渗均不破坏,冻干后重新以水悬浮并无明 显变化,pH<3.0时,可自然凝集,但pH达1.8时也不溶血;pH≥10.2可有中度溶血。醛化细胞悬液4可存放3个月以上,但冻结后重新融化则凝集成团。
 
戊二醛化红细胞制备法:
沉积红细胞用10~20倍0.11MpH7.2 PB洗4~6,再次配成10%悬液,取此悬液 (预先冷至4) 缓慢地加入到等量 1%的戊二醛(25%戊二醛用0.11MpH7.2PB稀释 之)中,并继续摇动30~60分钟,然后用蒸馏水洗五次。*后用重蒸馏水 配成20%的悬液,加0.1%NaN3防腐,4冰箱保存备用。
丙酮醛一一甲醛化红细胞制备:
(1)沉积红细胞以10~20倍体积 0.11MpH7.2PB洗涤4~6次,然后用该PB配成8%红细胞悬液。
(2)8%红细胞悬液中,缓慢加入等量3%丙酮醛、3%甲醛PB液(取丙酮醛15ml、甲醛8ml、0.11MpH7.2PB60ml,用10%Na-OH调正pH至7.2再加同样PB至100mI)于20左右缓慢搅拌17小时,至时用0.11MpH7.2PB洗三次,并用同样PB制成20%红细胞悬液,加入0.1%NaN3防腐,4冰箱保存备用。
注意事项:
1.在加醛类前要尽量将红细胞洗净;
2.开始加入甲醛浓度要淡,以后逐渐提高甲醛浓度,否则红细胞容易变形;
3.作适当的摇动,使甲醛与红细胞充分接触。
