二、实验材料与试剂配制：

1. 仪器与材料：酶标仪（使用前预热30分钟），微量加液器、吸头、蒸馏水或去离子水，滤纸。

2. 缓冲液使用：加5ul  的缓冲液于50ul  的样品中，如果样品量不够或者不确定，缓冲液和样品的混合比例不要小于1:10即可混匀，静置1小时备用（如果标本是血清或者血浆，此步骤忽略）。

3. 洗液的配制：按1:100的比例配制洗液备用。

1.检查试剂盒标签上的有效期。如果超过有效期，请勿使用。

2. 按说明书确定所有试剂齐全（数量、体积）

9.保证充分的孵育时间和温度。

1.在收集标本前都必须有一个完整的计划，必须清楚要检测的成份是否足够稳定。我们提倡新鲜标本尽早检测，对收集后当天就进行检测的标本，及时储存在4℃备用，如有特殊原因需要周期收集标本，请造模取材后，将标本及时分装后放在-20℃或-70℃条件下保存。因冰室与室温存在一定温差，蛋白极易降解，直接影响实验质量，所以避免反复冻融。部分标本也可加入适当防腐剂，激素类标本需添加抑肽酶。

3.液体类标本：标本必须为液体，不含沉淀。包括血清、血浆、尿液、胸腹水、脑脊液、细胞培养上清、组织匀浆等。
