二、实验材料与试剂配制：

1. 仪器与材料：酶标仪（使用前预热30分钟），微量加液器、吸头、蒸馏水或去离子水，滤纸。

2. 缓冲液使用：加5ul  的缓冲液于50ul  的样品中，如果样品量不够或者不确定，缓冲液和样品的混合比例不要小于1:10即可混匀，静置1小时备用（如果标本是血清或者血浆，此步骤忽略）。

3. 洗液的配制：按1:100的比例配制洗液备用。

1.检查试剂盒标签上的有效期。如果超过有效期，请勿使用。

2. 按说明书确定所有试剂齐全（数量、体积）

3. 标本制备要规范，每份标本体积要按2-3个复孔以上的量制备（贮存），尽量分装做备份。

4.短期内无法实验者，注意低温保存

5.准备好所有实验额外所需物品，（比如移液器，试管，清洗器，酶标仪）

6. 按说明书将所用试剂平衡至室温，根据检测标本数量确定所需试剂的量

8.检查不稳定或者变质的试剂溶液（比如沉淀或变色）。有些试剂，比如标准品稀释液或者检测稀释液，可能在设计时就含有沉淀物。所有试剂在滴入酶标板之前，必需充分混匀。而由于沉淀物的特性，在加入酶标板之前，必需不停搅拌。

9.保证充分的孵育时间和温度。

10.切勿使用不同生产批号的试剂取代现有试剂或混合现有试剂。

11.在混合或溶解蛋白溶液时，避免泡沫产生。

12.在实验开始之前，安排好实验流程。在实验开始之前，清洁工作台。

1.在收集标本前都必须有一个完整的计划，必须清楚要检测的成份是否足够稳定。我们提倡新鲜标本尽早检测，对收集后当天就进行检测的标本，及时储存在4℃备用，如有特殊原因需要周期收集标本，请造模取材后，将标本及时分装后放在-20℃或-70℃条件下保存。因冰室与室温存在一定温差，蛋白极易降解，直接影响实验质量，所以避免反复冻融。部分标本也可加入适当防腐剂，激素类标本需添加抑肽酶。 

2.试剂盒代测标本的客户取材前须向我司销售人员索要说明书，具体操作注意事项请与我司技术人员沟通。

3.液体类标本：标本必须为液体，不含沉淀。包括血清、血浆、尿液、胸腹水、脑脊液、细胞培养上清、组织匀浆等。

4.购买前请提前告诉我们：您样本的种属、样本数量、待测指标及其它特殊要求。
